Principleofthemethod
TheHumanAβ42USkitisasolidphasesandwichenzyme-linkedimmunosorbentassay(ELISA).AmonoclonalantibodyspecificfortheNH2-terminusofhumanAβhasbeencoatedontothewellsofthemicrotiterstripsprovided.Samples,includingstandardsofknownhumanAβcontent,controlspecimensandunknownsarePipettedintothesewells,andco-incubatedwitharabbitmonoclonalantibody*specificfortheCOOH-terminusofthe1-42Aβsequence.ThisCOOH-terminalsequenceiscreateduponcleavageoftheanalyzedprecursor.
BoundrabbitantibodyisdetectedbytheuseofahorserADIshperoxidase-labeledanti-rabbitantibody.Afterremovalofexcessanti-rabbitantibody,asubstratesolutionisadded,whichisacteduponbytheboundenzymetoproducecolor.TheintensityofthiscoloredproductisdirectlyproportionaltotheconcentrationofhumanAβ42presentintheoriginalspecimen.
Backgroundinformation
Alzheimer’sDisease(AD)ischaracterizedbythepresenceofextracellularplaquesandintracellularneurofibrillarytangles(NFTs)inthebrain.Themajorproteincomponentoftheseplaquesisβamyloidpeptide(Aβ),a40to43aminoacidpeptidecleavedfromamyloidprecursorproteinbyβ-secretase(i.e.,BACE)andaputativeγ(gamma)secretase.IncreasedreleaseofthelongerformsofAβpeptide,Aβ42orAβ43,whichhaveagreatertendencytoaggregatethanAβ40,occursinindividualsexpressingcertaingeneticmutations,expressingcertainApoEalleles,ormayinvolveother,stillundiscovered,factors.ManyresearcherstheorizethatitisthisincreasedreleaseofAβ42/Aβ43thatleadstotheabnormaldepositionofAβandtheassociatedneurotoxicityinthebrainsofaffectedindividuals.
